Sultan Sir Ismail mula menceburi sukan berkuda terutama polo ketika masih muda dan sering bersama ayahandanya menyertai pelbagai pertandingan. Baginda cekap dan mahir mengendalikan kuda sehingga berjaya melakar rekod kemenangan cemerlang setiap kali bertanding dalam pertandingan yang turut disertai oleh pelbagai negara luar termasuk Brunei dan Singapura.
Padang Polo di Pasir Pelangi pernah ditukar kepada Kelab Golf dan Tenis setelah penyertaan berkurangan sejak tahun 1933. Pada tahun 1950an baginda memperkenalkan Ismail Cup Polo Tournament yang diadakan di Singapura. Baginda menghidupkan kembali kegemilangan sukan polo Johor pada tahun 1961 dengan mengadakan pelbagai pertandingan jemputan antarabangsa.
Sultan Iskandar pula sering mendapat perhatian penonton kerana kecemerlangan baginda dalam pertandingan sukan polo dan aksi baginda disaksikan sendiri oleh ayahandanya dalam pertandingan-pertandingan yang disertainya. Lebih menarik, baginda mengetuai cabaran perlawanan pasukan polo dari Amerika Syarikat yang turut disertai oleh anakandanya Tunku Ibrahim disamping pemain berkelas tinggi dunia iaitu Sinclair Hill dari Australia dan Major Pradeep Mehra dari India pada tahun 1976 di Pasir Pelangi. Sultan Sir Ismail began developing an interest in horse riding especially polo from a young age and often spent time with His Majesty's father participating in various tournaments. His Majesty was proficient and skilled in handling horses until having an impressive winning record every time competing in any tournament that was also participated by other countries such as Brunei and Singapore.
The Polo Field in Pasir Pelangi was changed to the Golf and Tennis Club when the participation rate began to dwindle since 1933. In the 1950s, His Majesty introduced the Ismail Cup Polo Tournament that was held in Singapore. His Majesty brought back the glory days of Johor polo in 1961 by organising various international invitational tournaments.
Sultan Iskandar also attracted viewers' attention due to His Majesty's expertise in polo competitions and His Majesty's performances were witnessed by His Majesty's father in all competitions. Interestingly, His Majesty led a polo challenge from the United States that was also participated by son Tunku Ibrahim along with world class players Sinclair Hill from Australia and Major Pradeep Mehra from India in 1976 in Pasir Pelangi.
